Using homologous recombination strategy, two recombinase recognition sites (usually loxP) were introduced on each side of the target fragment individually. Since the recognition sites are usually located in introns that do not affect gene expression, mice exhibit a wild-type phenotype. Only in the presence of the recombinase (usually Cre), the target fragment is excised and the gene function is lost.
